Two people were killed and nearly 100 hit by bullets in the capital Sanaa, as police tried to prevent protesters marching towards the home of vice president Abd-Rabbu Mansour Hadi.
Others in the capital were injured by tear gas fired into the crowds and a further person was killed in the second city of Taiz.
The latest violent clashes came as Gulf mediators tried to broker an agreement between the opposition and government about a transfer of power from President Ali Abdullah Saleh.
Demonstrators threw stones at the anti-riot police and set fire to one security vehicle, witnesses said.
In Taiz, violence erupted as thousands more demonstrators also tested security forces' limits by burning piles of tyres and marching past the provincial governor's office, outside their usual protest area.
