The National Oganisation for Human Rights in Syria said 26 were killed in the southern city of Deraa and two in the central province of Homs.
A spokesman for the group said security and police forces had "dispersed peaceful gatherings in a number of Syrian provinces by using excessive and unjustified violence through the use of bullets".
It follows claims that 37 people were killed across the country on Friday.
"What is happening in Syria is a flagrant violation of (human rights)," the National Organisation for Human Rights said in a statement.
"Syrian security committed (in Deraa) what could be called a crime against humanity ... It fired indiscriminately on protesters and killed and wounded tens of them."
After Friday's violence, Syria's interior ministry warned it would not tolerate breaches of the law and would deal with "armed groups".
